ABSTRACT

Demyelination of the central nervous system often occurs in neurodegenerative diseases, such as multiple sclerosis (MS). The myelin sheath, a layer of myelin membrane wrapping the axon, plays a role in the rapid conduction and metabolic coupling of impulses for neurons. The exposure of the axon will lead to axonal degeneratio, and further neuronal degeneration, which is the main cause of dysfunction and even disability in patients with demyelinating neurodegenerative diseases. In addition to the demyelination of mature myelin sheath, remyelination disorder is also one of the major reasons leading to the development of the diseases. The myelin sheath is composed of oligodendrocytes (OLs) derived from oligodendrocyte progenitor cells (OPCs) which are differentiated from neural stem cells (NSCs). The process of myelin regeneration, i.e., remyelination, is the differentiation of NSCs into OLs. Recent studies have shown that this process is regulated by a variety of genes. MicroRNAs, as important regulators of neurodegenerative diseases, form a complex regulatory network in the process of myelin regeneration. This review summarizes the main molecular pathways of myelin regeneration and microRNAs involved in this process and classifies the mechanisms and targets. This review is expected to provide a theoretical reference for the future research on the treatment of demyelinating diseases by targeting the regulation of microRNAs.

ABSTRACT

ObjectiveTo observe the protective effect of Shenlian prescription on acute lung injury induced by particulate matter (PM) exposure in rats and explore the mechanism. MethodFifty male SD rats were randomly divided into the control group, model group, Shenlian low-dose group (4.32 g·kg-1), Shenlian high-dose group (8.64 g·kg-1), and roflumilast group (3.46 mg·kg-1), with 10 in each group. Pre-administration with drugs by gavage was performed for one week. On the 8th and 11th days, the control group was instilled with normal saline in the trachea and the other groups with PM suspension to establish a rat model of acute lung injury induced by PM exposure. After modeling, drugs were given continuously until the end of the experiment. Forty-eight hours after the last exposure, the lung function of rats was detected. Then the rats were sacrificed and the lung morphological changes and pathological changes by hematoxylin-eosin (HE) staining were observed. CD68 expression in lung was detected by immunohistochemistry, and the levels of lung injury markers surfactant protein A (SP-A) and Clara cell protein16 (CC16) in serum were detected by enzyme-linked immunosorbent assay (ELISA). The mRNA expression of interleukin-1α (IL-1α), IL-6, IL-18, and monocyte chemoattractant protein-1 (MCP-1) in lung tissue was measured by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). ResultCompared with those in the control group, the rats in the model group had decreased lung function and obvious structural damage of lung tissue, PM deposition, and infiltration of CD68 positive cells. The expressions of IL-1α, IL-6, IL-18, and MCP-1 in lung tissue were increased (P<0.01). Compared with the model group, Shenlian prescription low and high doses restored the rats' lung function injury(P<0.05,P<0.01), improved lung morphological and pathological structure, and reduced PM deposition. Infiltration of CD68 positive cells in lung was not significantly decreased. The levels of inflammatory factors IL-1α, IL -6, IL-18, and MCP-1 in lung were lowered (P<0.01). ConclusionShenlian prescription could protect the rats' lung injury caused by PM exposure, improve lung morphology, and reduce PM deposition and inflammatory factor expression.

ABSTRACT

ObjectiveTo explore the effects of anxiety and depression on Internet gaming disorder in medical students, and to evaluate the moderating effect of gender, so as to provide references for the prevention and intervention of Internet gaming disorder in medical students. MethodsIn November 2021, a total of 11 771 medical students from a medical college in Sichuan province participated in an online survey through Wenjuanxing platform. Participants completed the Self-rating Anxiety Scale (SAS), Self-rating Depression Scale (SDS) and the nine-item Internet Gaming Disorder Scale-Short Form (IGDS9-SF). Then a hierarchical multiple regression analysis was conducted to discuss the moderating role of gender in the relationships between anxiety, depression and Internet gaming disorder in medical students. Results①Males scored lower on SDS (t=-8.302, P<0.01), and higher on IGDS9-SF than females (t=33.384, P<0.01). ②SAS score of medical students was positively correlated with SDS score (r=0.735, P<0.01), SAS and SDS scores were positively correlated with IGDS9-SF score in medical students (r=0.288, 0.238, P<0.01). ③Anxiety and depression could positively predict Internet gaming disorder (β=0.245, 0.058, t=18.864, 4.444, P<0.01). ④Gender played a moderating effect between anxiety and Internet gaming disorder (β=-0.194, t=-4.518, P<0.01). ConclusionAnxiety and depression have a positive predictive effect on Internet gaming disorder in medical students. Furthermore, the effects of anxiety on Internet gaming disorder are moderated by gender, and the anxiety of male students exhibits a markedly stronger impact on Internet gaming disorder than that of female students.

ABSTRACT

OBJECTIVE:To study the sta bility,in vivo release characteristics and tissue distribution of docetaxel (DTX)- dihydroartemisinin(DHA)conjugated prodrug self-assembled nanoparticles (DTX-S-S-DHA NPs ). METHODS :HPLC method was adopted to analyze DTX-S-S-DHA in vitro . The phycial and long-term stability of DTX-S-S-DHA NPs in mediums [water , saline, phosphate buffer (PBS,pH 7.4)and RPMI 1640 medium] were investigated by using particle size ,polydispersity index (PDI)and encapsulation efficiency (EE)as evaluation indexes. The in vitro release characteristics of DTX-S-S-DHA released from DTX-S-S-DHA NPs was also investigated with small glass method ,using 30% ethanol solution with or without 10 mmol/L dithiothreitol(DTT)as medium. The small live animal imager was adopted to investigate the tissue distribution and tumor targeting capability of DiR-labeled DTX-S-S-DHA NPs (DTX-S-S-DHA/DiR NPs )in breast cancer bearing mice. RESULTS :In stability test, there was no statistical difference in particle size ,PDI and EE of DTX-S-S-DHA NPs incubated in water ,normal saline ,PBS and RPMI 1640 medium for 24 h. When stored at 4 ℃,with the increase of storage time ,the particle size of DTX-S-S-DHA NPs in normal saline gradually increased ,while those in PBS gradually decreased ;EE of both gradually decreased to less than 75%, but there was no significant change in particle size ,PDI and EE of DTX-S-S-DHA NPs in water and RPMI 1640 medium. In the in vitro release experiments ,DTX-S-S-DHA in DTX-S-S-DHA NPs was not released in the release medium containing 10 mmol/L DTT;at 24 h,the cumulative release rate of DTX-S-S-DHA released from DTX-S-S-DHA NPs in release medium without DTT was about 83%,which was in line with first-order kinetic model. In tissue distribution test ,the distribution of DTX-S-S-DHA/DiR NPs in tumor sites of mice was significantly more than in other tissues (heart, liver, spleen, lung and kidney ). CONCLUSIONS : DTX-S-S-DHA NPs show good physical stability in different mediums ,especially have good long-term stability in water and RPMI ; 1640 medium;they can quickly release the parent drug in the reduction environment and has good tumor targeting.

ABSTRACT

Objective@#To compare the effect on the flow conditions of adhesives and the retention force of restorations among different cement-retained methods of implant-supported fixed prostheses.@*Methods@#Four common cement-retained methods were selected, including the occlusal hole for screw access (OH), the lingual hole for adhesives overflow (LH), the resin replica for titanium abutment (RR), and the traditional cement-retained method (the control group). The adhesive used in this study was resin-modified glass ionomer cement. The two-dimensional analysis models of computational fluid dynamics (CFD) were established. The flow conditions of adhesives in the adhesion process was analyzed by the CFD analysis. The internal filling ratio and the amount of neck overflow of adhesives below the edge of the prosthesis were calculated. Ten zirconia prostheses in each group were processed and cemented. The retention force was examined by mechanical tensile experiments in vitro.@*Results@#The CFD analysis showed the internal filling ratio of adhesives from high to low was the LH group, the OH group, the RR group and the control group. The amount of neck overflow of adhesives below the edge of the prosthesis from less to more was the RR group, the OH group, the LH group and the control group. The retention force was (240.7±33.9) N in the control group, (278.2±59.1) N in the OH group, (292.9±47.9) N in the LH group, and (262.8±59.4) N in the RR group. There was a statistically significant difference in the retention force between the LH group and the control group (P=0.029). There was no significant difference among the other groups (P>0.05).@*Conclusions@#The modified cement-retained methods can effectively reduce the amount of neck overflow of adhesives, and improve the filling condition of adhesive in the adhesive clearance to ensure the retention force of the prostheses. Clinically, the appropriate modified cement-retained method should be selected according to the three-dimensional position of the implant and the position of prosthetic margin.

ABSTRACT

The proliferation of tumor cells is regulated by a complex array of signaling pathways, among these signaling pathways, the programmed cell death. Autophagy and apoptosis are two types of programmed death. There are significant differences in their morphological and functional features, but they also have many links. Both apoptosis and autophagy are involved in activation, expression and regulation of a series of genes. By reviewing the research progress in recent years, this article will discuss the cellular regulation and molecular mechanisms of their related genes. Through summarizing the relationship between autophagy and apoptosis,it aims to get a better understanding of the mechanisms of autophagy and apoptosis in tumor progression, and looking at the perspectives for studies on the autophagy and apoptosis in tumor treatment.

ABSTRACT

Objective To explore the biological indicators of diagnosis and treatment of irritable bowel syndrome (IBS), and to explore the mechanism of action of a Chinese medicine Wuji Pill (WJW) on irritable bowel syndrome (IBS). Methods (1) Postinflammatory irritable bowel syndrome (PI-IBS) rat model was established by acetic acid plus restraint stress method . (2) The colonic motor ability of rats was evaluated by colon motility index (MI), the number of fecal particles discharged within 2 h, and the time of glass pellet discharge. (3) The formation of PI-IBS model rats and the therapeutic effect of WJW were observed. (4) The levels of calcitonin gene-related peptide (CGRP), motilin (MTL), neuropeptide Y (NPY), substance P (SP), somatostatin (SS), vasoactive intestinal peptide (VIP), and cholecystokinin (CCK) in the brain and colon tissues of PI-IBS rats were measured by ELISA. Results (1) The rat PI-IBS model was successfully established. Compared with the normal group, the body weight of the model rats was decreased, the food intake decreased, the amount of feces increased, loose stools and amorphous soft stools appeared, voluntary movements decreased, colon motility index ( MI) significantly increased ( P < 0. 05 ), the number of fecal particles discharged significantly increased ( P< 0. 05), and the glass pellet discharge time was significantly shortened ( P < 0. 05). (2) WJW treatment for 7 days significantly improved a variety of symptoms. Compared with the normal control, the levels of CGRP, SS and VIP in the brain tissue of PI-IBS rats were significantly increased (P< 0. 05), and the NPY concentration was significantly decreased ( P < 0. 05). However, the treatment with WJW significantly reduced CGRP, SS and VIP levels (P< 0. 05), and significantly increased the NPY concentration level (P < 0. 05). (3) Compared with the normal control group, the levels of CCK, NPY, MTL, SS and VIP in colonic tissues of PI-IBS rats were significantly decreased (P< 0. 05), while WJW significantly increased the CCK and VIP levels. Conclusions WJW can be used to treat IBS by regulating the levels of various brain-gut peptides in the brain and colon tissues of IBS rats. These anomalous and adjustable brain-gut peptides may become a potential biomarker for the diagnosis and treatment of IBS.

Neurodegenerative diseases are threating our health seriously.Inflammation plays an important role in the initiation and development of neurodegenerative diseases, and its primary characteristics are the activation of microglia and the increasing level of inflammation cytokines. This review describes the relationship between neuroinflammation and several neurodegenerative diseases, and the models in vivo and in vitro. In addition, combining with traditional Chinese medicines knowledge of encephalopathy, we summarizes pharmacological effects and mechanisms of multiple herb extracts and monomer compounds in preventing the activation of microglia and inhibiting neuroinflammation, thus, to provide the basis for gradually revealing the related rules and characteristics of treating encephalopathy by traditional Chinese medicine, and improving the accuracy of the clinical drugs, as well as developing new drugs for the prevention and control of encephalopathy.

ABSTRACT

Objective To investigate the effects of Siwu decoction on the learning and memory abilities in mice with blood deficiency and the related mechanism. Methods The mice were randomly divided into five groups:normal control group, model control group,high-,medium-,and low-dose group of Siwu decoction.Blood deficiency mouse model was established by continuously cuttingtailand bleedingin model control group,high-,medium-,and low-dose group of Siwu decoction. The mice in high-,medium-,and low-dose group were intragastrically administrated with Siwu decoction of 2.5,5.0,10.0 g·kg-1 for 14 days, respectively. The erythrocyte counts (RBC),hemoglobin (HGB),red blood cell hematocrit (HCT) were detected before modeling,7 and 14 days after administration. The spatial learning and memory abilities were assessed using the T maze test. The spontaneous activities were assessed using locomotors activity detector. The levels of erythropoietin (EPO) content in serum and acetylcholinesterase (AChE) in hippocampal tissue were detected by enzyme-linked immunousorbent assay. Results 5.0 and 10.0 g·kg-1Siwu decoction could increase RBC,HGB,HCT on the 7th and 14th day,2.5 g.kg-1Siwu decoction could increase RBC,HCT on 14th day,and each dosage of Siwu decoction could significantly increase the spontaneous activities of model mice with blood deficiency on the 7th and 14th day as compared with model control group (P <0.05 or P <0.01).Compared with model control group, the number of errorsin T Maze test were decreased significantly in high-,medium-,and low-dose groups (P < 0.01).And the Siwu decoction had a tendency to reduce the hippocampal AChE levels, but when compared with the model control group there were no statistical differences (P > 0.05).Conclusion Siwu decoction is benefit formemory and spatial learning in mice with blood deficiency, which may be related with higher serun EPO and lower hippocampal AChE expression.

ABSTRACT

Objective To assess the diagnostic value of combining aortic dissection detection risk score (ADDRS)with D-dimer in patients with suspected acute aortic dissection(AAD). Methods We retrospectively identified 525 patients with chest pain and with calculated ADDRS and available D-dimer on admission between January 2015 and December 2016. ADDRS ≤1 was defined as ′low probability′ and that >1 as ′high probability′. Results AAD were diagnosed in 106(20.2%)patients. In patients with ADDRS of 0,1 case of AAD was identi-fied and in those with ADDRS of 1,40 were identified while in 76 patients with ADDRS of 2 and 3,71 cases of AAD were identified. D-dimer with threshold of 0.5 mg/L had a negative predictive value of 100%and a specificity of 76.6% in patients with ADDRS of 0. In patients with ADDRS of 1,D-dimer had a negative predictive value of 98.6%. D-dimer showed a negative predictive value of 99.6% and a positive predictive value of 22.3% in patients with low probability. Conclusions High ADDRS benefits confirming AAD. Low ADDRS combined with negative D-dimer is safe and efficient for ruling out AAD ,while low ADDRS with positive D-dimer needs further aortic imaging.
